The library of the Mainz Carthusian monastery, which was dissolved in 1781, is the most important source of the mainly late medieval manuscripts. Other sources include the Carmelites, the Benedictine monastery of St. Jakob, the Jesuit college, the Capuchins, the Augustinian hermits, and the old University of Mainz.

The old holdings are available for use in the first row of the reading room. This is based on the "Special provisions for the use of manuscripts, rare books, and special collections."
